Zebularine

ACCESSION NB: DB03068 (EXPT03280)

TYPE: small molecule

GROUP: experimental

DESCRIPTION:
A chemically stable, cytidine analog that displays anti-tumor properties. Acts as a transition state analog inhibitor of cytidine deaminase by binding to the active size as covalent hydrates. Also shown to inhibit DNA methylation and tumor growth both in vitro and in vivo.

MECHANISM OF ACTION:
Inhibition of cytidine deaminase by zebularine enhances the antineoplastic action of 5-aza-2'-deoxycytidine. Cytidine (CR) deaminase is a key enzyme in the catabolism of cytosine nucleoside analogues, since their deamination results in a loss of their pharmacological activity. Zebularine has a dual mechanism of action, since it can also inhibit cytidine (CR) deaminase.
